ABSTRACT

Objective To explore the clinicopathlogical characteristics and prognostic factors of male breast cancer.Methods Clinical data from 30 cases of male breast cancer patients were collected, the survival curve of PFS was drawn and the survival rate and survival prognosis were estimated.Results There were 29 cases(97%)of estrogen receptor positive patients in total. The tumor pathology grades (χ2=6.141,P=0.013)and the status of hormone receptor(χ2=10.465,P=0.005)affected the prog-nosis of male patients with breast cancer in univariate analysis. And the tumor pathology grades(χ2=5.271,P=0.022,HR =5.762)affected the prognosis in multivariate analysis. Conclusion In male breast cancer patients,the positive rate of estrogen receptor is relatively high,and the tumor pathology grades is one of the most important factors affecting prognosis.

ABSTRACT

Objective To evaluate the roles of signal transducer and activator of transcription 3(STAT3) played in delayed xenograft rejection.Method Mice-to-rats cardiac xenograft model was established and recipients were administrated by inhibitor of phosphorylation of STAT3,5,15-Diphenylporphyrin(DPP),and immunosuppressive agent,ciclosporin A (CsA) alone or both.The survival of graft was analyzed.Immunohistochemistry and immunoblotting were utilized to detect the phosphorylated STAT3 (p-STAT3),and real-time polymerase chain reaction was used to assess STAT3-targeted genes in grafts.Result The expression of p-STAT3 was increased significantly with the prolonged survival in grafts (P < 0.05).Administration of DPP and CsA both significantly prolonged survival of the grafts (P<0.05),and decreased the expressions of STAT3-targeted genes including Bcl-xl,Bcl-2,Cyclin D1,C-myc and VEGF (P<0.05).DPP and CsA exerted the synergistic effects.Conclusion STAT3,which is persistently activated in cardiac xenografts,probably up-regulates downstream genes to promote proliferation and to inhibit apoptosis of endothelial cells,leading to the activation of endothelial cells and delayed xenograft rejection.

ABSTRACT

<p><b>OBJECTIVE</b>To investigate the therapeutic effects of closed reduction and external fixation (plaster or splint) for the treatment of displaced humeral supracondylar fractures in children.</p><p><b>METHODS</b>From March 2007 to September 2009,33 children (15 female and 18 male) with humeral supracondylar fractures treated in our hospital, ranging from 3 to 12 years old with an average of 6.4 years old. All the fractures were extension-type injuries, the flexion injures were excluded in our study. The humeral supracondylar fractures were classified according to Gartland classification. There were 21 Type H and 12 type III. In the initial treatment, all the patients were treated with closed reduction and external immobilization. The blood supply of the damaged upper extremity was evaluated before and after treatment. Clinical assessment was obtained at final follow-up using Flynn criteria, and radiologic assessment was obtained using Baumann and lateral humerocapitellar angles.</p><p><b>RESULTS</b>All the children were treated successfully with closed reduction in the initial time; 24 children maintained limb alignment by external immobilization. Nine patients lost position due to the swelling around the elbow which affected unstable external fixation during the follow-up, 5 of which were treated with a repeated closed reduction and internal fixation with Kirschner wires, 4 of which were treated with traction. Thirty-one patients had a satisfactory outcome and 2 patients had an unsatisfactory outcome according to the Flynn criteria at the latest follows-up.</p><p><b>CONCLUSION</b>Closed reduction and external stabilization is an important method for the treatment of displaced humeral supracondylar fractures in children. Making regular follow-up visits after closed reduction and casting is important for patients to maintain acceptable alignment, avoid complications and diagnose any loss of reduction.</p>

ABSTRACT

PURPOSE: G-protein coupled estrogen receptor 1 (GPER) probably play important roles in the progression of breast cancer including endocrine therapeutic resistance. We evaluated GPER in primary breast cancers. METHODS: Immunohistochemistry was used to detect GPER in paraffin-embedded tissues of primary breast cancers from 423 patients and GPER expression was correlated with clinicopathological factors. RESULTS: GPER was expressed in 63.8% of specimens, coexpressed with estrogen receptor alpha (ERalpha) in 36.6% of tumors and was positive in 62.5% of the ERalpha-negative tumors. The expression of GPER had no relationship with the status of ERalpha, progesterone receptor and HER2. Although the expression of GPER was significantly inversely related with nodal status (p=0.045), no correlation between GPER expression and other clinicopathological variables (age, menstruation status, tumor size, stage, histologic grade, Nottingham Prognostic Index or pathological type) was found. CONCLUSION: GPER and ERalpha exhibited independent expression pattern of distribution in primary breast cancers. A long-term follow-up and a more definite molecular phenotype for ER are necessary in confirming studies.

ABSTRACT

Objective Comparing the influence of neoadjuvant chemotherapy on sentinel lymph node biopsy and axillary lymph nodal staging. Methods From June 2005 to June 2007 one hundred and sixty five breast cancer patients of stage Ⅰ,Ⅱa,Ⅱb and Ⅲ underwent sentinel lymph node biopsy(SLNB) using 99mTc combining methylene blue dye as tracer to help axillary lymph node dissection(AND) were retrospectively reviewed. Sixty five patients staged Ⅱb or Ⅲ were treated with three circles neoadjuvant chemotherapy prior to SLNB and AND , another one hundred patients staged ⅠorⅡa had SLNB and AND directly without neoadjuvant chemotherapy.Results Average 14.60 axillary lymph nodes were retrieved in patients had neoadjuvant chemotherapy and 14.74 lymph nodes in patients did not have neoadjuvant chemotherapy(P>0.05),a mean number of 1.46 sentinel lymph nodes in neoadjuvant chemotherapy group and 1.5 sentinel lymph nodes in non-neoadjuvant chemotherapy(P>0.05), sentinel lymph node identification rate 96.9% in neoadjuvant chemotherapy group and 97% in non-neoadjuvant chemotherapy(P>0.05), the false negative rate was 4.6% in neoadjuvant chemotherapy group and 4% in non-neoadjuvant chemotherapy group(P>0.05). Conclusions The mean number of lymph nodes,sentinel lymph nodes,SLN identification rate and false-negative rates after neoadjuvant chemotherapy are similar to those seen in patients without neoadjuvant chemotherapy.The SLNB can accurately predict lymph node status of axillary lymph node following neoadjuvant chemotherapy.

ABSTRACT

Objective To study expression enhancement and significance of Y14 and Upf1 in human breast cancer cell lines and tissue. Methods Immuocytochemistry and laser scanning confocal microscope(LSCM) were applied. Y14 and Upf1 were determined in human breast cancer cell lines(MCF-7,ZR-75-30,T47D,MDA-MB435s,MDA-MB-453, MDA-MB-231) and breast epithelial cell line ( HBL-100). Results (1) Y14 and Upf1 level of breast cancer cells are obviously higher than that in breast epithelial cell line (P < 0. 05 ). (2)Y14 and Upf1 level of MDA-MB-231 are obviously higher than that in MCF-7. (3)The expression enhancement of Y14 and Upf1 level are obviously higher in human breast cancer tissue. Conclusion The expression level of Y14 and Upf1 in breast cancer cells and tissue enhance obviously.

ABSTRACT

G protein-coupled receptor 30(GPR30) was a novel estrogen receptor identified as membrane associated receptor in the late 1990s.This new member of estrogen receptors was independent of the classic nuclear estrogen receptor ? and ? due to the low homology and signifi cant difference between them.It was reported that GPR30 localized endoplasmic reticulum predominantly,which was expressed in diverse cancer cells and a wide range of systems throughout the body.The rapid non-genetic response,partially at least,transcription regulation of estrogenic effects were mediated by the novel receptor via transactivation of epidermal growth factor receptor and modulation of second messengers such as cyclic adenosine monophosphate(cAMP) and Ca2+.These pathways,possibly coordinate with ER?,were involved in various physiological,physiopathological and carcinogenesis process.Theoretically,GPR30 would be a novel therapeutic target in estrogen-related diseases such as breast carcinoma.

ABSTRACT

Objective To denoise digital radiographic images well.Methods A technique was presented that used the Anscombe's transformation to adjust the original image to a Gaussian noise model based upon the wavelet denoising method and the wavelet-domain Hidden Markov Tree(HMT) model.Wavelet domain HMT models were used to determine the dependencies of multiscale wavelet coefficients through the state probabilities of the wavelet coefficients,whose sedistribution densities could be approximated by Gaussian mixture model.Results The proposed method could keep natural images edges from damaging and increase PSNR.Conclusion Quantitative and qualitative DR images assessment shows that the proposed algorithm outperforms the traditional Gaussian filter in terms of noise reduction,quality of details and bone sharpness.
